Djokovic routs Federer, faces Nadal for title

For the fourth straight time in a Grand Slam final it will be Novak Djokovic vs. Rafael Nadal.? That Nadal won his French Open semifinal in a breeze against David Ferrer was no shock.? That Djokovic ran into only a wisp of a challenge from Roger Federer ? well, that came as a bigger surprise.

Orioles win at Fenway for 7th straight time

Associated Press Sports

updated 10:16 p.m. ET June 6, 2012

BOSTON (AP) - Baltimore manager Buck Showalter decided it was time to let Wei-Yin Chen handle things on his own a little more.

The rookie left-hander passed the test.

Chen scattered seven hits over seven innings and the Orioles set a franchise record with their seventh straight victory in Boston, beating the Red Sox 2-1 on Wednesday night.

Chen's most important inning was the seventh, when Boston had runners on second and third after a pair of singles and a sacrifice. He struck out Marlon Byrd and got Mike Aviles to pop up to short right, escaping the jam.

"He'd pitched too effectively not to let him decide his own fate," Showalter said.

Chen, the first Taiwanese-born player in Orioles history, is slowly building a nice season. He understood the faith his manager showed.

"I really appreciate that Buck trusted me," Chen said through a translator. "That's the key point."

And the Orioles, who moved into sole possession of first place in the AL East, made another point that they're proving to be trouble for the Red Sox again this season.

The Orioles, who knocked Boston out of the postseason on the final night of the regular season last year, have won 12 of the past 15 meetings. They are 5-0 at Fenway Park this season.

Endy Chavez, who had three hits in a series-opening win Tuesday, drove in the go-ahead run with a grounder in the sixth. Boston has lost three straight for the first time since early May.

"Everybody's doing a little part for the team," Chavez said. "As a team, we are playing pretty good even though we have a lot of injuries. We're playing good. We're focused and we're fighting and all together. That's very important."

Chen (5-2) was coming off the worst start of his first major league season, when he allowed five first-inning runs Friday in a loss to Tampa Bay. This time he struck out four, didn't walk anyone and stranded six runners.

Josh Beckett (4-6) was the tough-luck loser, allowing two runs, five hits and striking out five in eight innings. It was the fifth consecutive start he's pitched at least seven innings.

"I learned a lot from him tonight," Chen said of going against Beckett. "I haven't had a win in a long time. It was a relief to me."

Chen went 0-2 with a no-decision in his previous three starts.

Aviles thought Boston's best shot came in the seventh.

"Yeah, that's exactly what it came down to. Their pitchers basically outpitched our hitters," he said. "It was a great-pitched ballgame all around. Josh pitched unbelievably well and he definitely deserved to win and unfortunately we weren't able to back him up with the bats."

Beckett declined to talk to reporters.

Jim Johnson, who had his first blown save of the season Tuesday after converting 25 straight chances dating to last year, got three outs for his 18th save. The first batter he retired was Jarrod Saltalamacchia, who hit a tying, two-run homer one night earlier.

Trailing 1-0 and with just a single in the first five innings, the Orioles opened the sixth with three consecutive hits and took the lead with two runs. Robert Andino had an RBI single after Wilson Betemit and Ryan Flaherty singled. Chavez's fielder's choice made it 2-1.

The Red Sox grabbed a 1-0 lead in the third when Darnell McDonald doubled off the Green Monster, advanced on Marlon Byrd's single and scored on Mike Aviles' sacrifice fly.

Chen allowed hits in every inning except the first, but kept the damage to a minimum.

With help from third baseman Will Middlebrooks, who made a diving grab of Mark Reynolds' second-inning liner, Beckett retired the first nine batters on 29 pitches, fanning three.

Beckett faced the minimum and held the Orioles to one runner through the first five innings - Chavez's single leading off the fourth. Chavez was erased when J.J. Hardy bounced into a double play.

NOTES: The Orioles signed 49-year-old LHP Jamie Moyer to a minor league deal and assigned him to Triple-A Norfolk. He is expected to start Saturday against Buffalo. Showalter said Moyer will get "two or three starts down there and we'll see what happens." Moyer became the oldest pitcher to win as a starter when he earned a victory with Colorado in April. ... Red Sox manager Bobby Valentine said RHP Daisuke Matsuzaka will start Saturday against Washington, one day before the one-year anniversary of his Tommy John surgery. Matsuzaka made eight rehab starts and is taking the spot of Daniel Bard, sent to Triple-A Pawtucket on Tuesday. Bard, a converted reliever, is 5-6 with a 5.24 ERA. ... The Orioles selected Cal Ripken Jr.'s son, Ryan, in the 20th round of the draft. "That was pretty cool," Showalter said. "This kid's got a chance to be a player. He's an outstanding prospect." The 6-foot-6 Ryan Ripken played at the Gilmon School in Baltimore. ... Showalter also said he heard 2B Brian Roberts, attempting to come back from a concussion sustained more than a year ago, "looked good. He's moving around and playing good defense." Roberts is playing in the minors.

Taylor Swift's Label Inks Groundbreaking Deal With Clear Channel

Artists on Big Machine Records will now receive payment every time their songs are played on Clear Channel radio stations.

Taylor Swift is in a brand-new relationship. No, she isn't dating some hot Hollywood star. Instead, Swift's label, Big Machine Records, has just inked a deal with Clear Channel that will allow artists on the label to get paid for songs played on their radio stations.

The label, as well as the performers on it — including heavy hitters like Tim McGraw, Rascal Flatts and Reba McEntire — will share the revenue with Clear Channel. Before this deal, the label and its performers did not collect royalties when their songs were played on terrestrial radio; only songwriters and publishers did.

Internet radio stations, however, already do pay performers and labels when their songs are played. Under the new deal, Clear Channel will be able to limit payments for songs streamed online, which will enable the growth of digital radio.

"Not only does this partnership enable Big Machine to participate in terrestrial broadcast revenues, but we are also helping to grow digital radio," Scott Borchetta, the label's president and CEO, said. "[It's] a great opportunity for all of us and a breakthrough opportunity for Big Machine artists."

But reportedly, it won't end there. Aceshowbiz.com further reports that Clear Channel will approach more labels for similar deals and Big Machine may ink more of these with other radio conglomerates.

"Today, 98 percent of our listening is terrestrial broadcast and 2 percent digital — with record labels and artists only paid for the 2 percent," John Hogan, chairman and CEO of Clear Channel Media and Entertainment, added in a statement about the landmark deal. "This new agreement expands label and artist participation from just digital to terrestrial broadcast radio revenue in one comprehensive framework that will give all of us a great incentive to drive the growth of the digital radio industry and allow everyone to participate financially in its growth."

This coup for Swift comes just months after she was named the top earner of 2012 by Billboard magazine on their "Music's Top 40 Money Makers" list. Swift sits at #1 thanks to reported earnings of $35.7 million in the 2011 calendar year. She ranks so high due to quadruple-platinum-plus sales of Speak Now and its accompanying tour, which grossed $88.5 million in the U.S. alone.

But she's hardly resting on her laurels. She is currently hard at work on her follow-up to the release. There is no word yet on when it might drop.

Thunder reaches NBA Finals by beating Spurs in Game 6

For the first time since 1996, when the franchise was in Seattle, Oklahoma City is in the NBA Finals by virtue of their series-clinching win Wednesday night over San Antonio.

"That's his first charge of the year," Russell Westbrook interjected when Durant was asked about drawing an offensive foul against Manu Ginobili in the fourth quarter of Oklahoma City's 107-99 win in Game 6 of the Western Conference finals on Wednesday night.

Indeed, it was.

The league's three-time scoring champion had 34 points and 14 rebounds while playing all of regulation for the first time all season, leading the Thunder into the NBA finals. But it was taking that charge that got his team pumped up.

Durant stepped in front of Ginobili's drive during a 3?-minute scoreless stretch by San Antonio that allowed Oklahoma City to take the lead for good.

"I just wanted to go out there and sacrifice my body for my team. I knew that would give us a little spark," Durant said.

"Manu's an unbelievable player at twisting his body and making crazy shots, so I just wanted to time it right. It felt good to get that for my team and I could tell they were excited that I got my first one when I looked at the bench."

Westbrook added 25 points for the Thunder, who trailed by 18 in the first half and erased a 15-point halftime deficit.

The Thunder took the lead for good early in the fourth quarter, getting nine of their first 13 points on free throws as the fouls started to pile up for San Antonio ? six on the defensive end and three on the offensive end in the first 7 minutes.

That included Durant's stop just outside the restricted area under the basket.

"Down the stretch, it seemed like they got every whistle possible and that really changed the tide," San Antonio's Tim Duncan said. "We were playing tough defense and trying to get stops, but the whistle kept blowing and they went to the line."

Tony Parker finished with 29 points and 12 assists, but only eight of the points and two assists came after San Antonio took a 63-48 halftime lead. Duncan chipped in 25 points and 14 rebounds, and Stephen Jackson hit six 3-pointers and scored 23 points.

The Spurs had won 20 in a row, moving past the Thunder for home-court advantage in the West and then taking a 2-0 lead in the series, before losing four in a row.

"There's not much to complain about," Ginobili said. "We had a great run. We just couldn't beat these guys."

Durant grabbed the final rebound, dribbled the ball across half court and raised his right fist to celebrate with a sold-out crowd wearing free white T-shirts. The franchise will play for the NBA title for the first time since 1996, when it was in Seattle.

Game 1 of the NBA finals will be Tuesday night in Oklahoma City against either Boston or Miami. The Celtics lead that series 3-2 and can earn a trip to the finals with a win at home in Game 6 on Thursday night.

Preparing for a Workers' Compensation Audit | Oyer, Macoviak, and ...

Just the mention of a workers? compensation audit engenders fear into the heart of man. If you are scheduled for a workers? compensation audit, there is no need to dread it or be fearful. A small amount of preparation and common sense can save you a lot of aggravation and money.

Devoting a few hours of your attention now can save you a lot of time in the future. You need to give your full attention to the auditor and remain with him/her throughout the entire process. The process may take a couple of hours to complete. So, make sure that the time and date of the scheduled audit is convenient for you. Call to reschedule the audit if it has been scheduled at an inopportune time.

Begin to collect and organize payroll records, overtime payroll records, classification divisions, and insurance certificates as soon as you learn of the audit. This should give you plenty of time to compose a summary of each, which will help you to better communicate important data during the audit process. Information that is well organized will also expedite the process. If you can reconcile your calculations to payroll records, such as W2?s and payroll stubs, the auditor might be more comfortable trusting your data.

You will also want to make any needed adjustment to payrolls; for example, subtracting bonus pay from overtime pay. If applicable, you will need to apply the maximum and minimum payrolls to the calculations. This part may take a little research, since the minimum and maximum will vary based on state; career; and even among sole proprietors, partners, and executive officers.

Before the audit, you should apprise yourself on the different employee job classifications and make sure that each employee is correctly classified. This is a key element to ensure that the audit flows smoothly. If there is any question about how to classify an employee, call your broker or agent. The auditor is most likely going to be asking you about the classification and job duties for multiple different employees. So, be prepared and armed with knowledge.

One last important preparation is concerning subcontractors. Payments issued to subcontractors can go against your workers? compensation in the event that the subcontractor did not have a certificate of workers? compensation. You can get a copy of the certificate, but make sure that it is current and shows coverage during the time the subcontractor worked for you.

Once the audit arrives, you will be glad that you took a little time collecting, organizing, and summarizing your information. You will also find that the auditor is not a nemeses, especially when you provide honest answers and organized paperwork.

At the conclusion of the audit, ask the auditor for the audit worksheet. Ask your broker or agent to review the accuracy of the final audit. You have a legal right to ask for a corrected audit anytime you think there were any errors. If any overpayment was made under the last three preceding audits, you also have a legal right to recover it.

FDA to let women try new breast drugs earlier

CHICAGO (Reuters) - Regulators are moving the goal posts in testing new drugs for breast cancer in the hopes of giving more women with aggressive, early-stage cancers the chance to try breakthrough drugs while they have the best shot at a cure.

A new guidance document issued this week by the U.S. Food and Drug Administration will allow drug companies to test their medications for a few months on women with highly aggressive breast cancers before they have surgery, instead of waiting until the drug has been proven first in gravely ill patients.

"We're looking at introducing drugs into a very early stage of breast cancer, where a patient has a primary tumor and the chemotherapy is given before surgery," Dr. Richard Pazdur, director of the FDA's cancer drug office, said in an interview at the American Society of Clinical Oncology meeting in Chicago.

"The aim of this therapy would be to cure the woman of breast cancer," Pazdur said. "It's moving a very promising drug into a early stage of the disease with a curative intent."

If a drug succeeds in eradicating the cancer to the point where there is no sign of it in the breast or lymph nodes - something called pathologic complete response - companies could win accelerated approval.

That represents a significant change from the current approach, in which promising drugs only are tested in earlier stage cancers after they have first proven to be safe and effective in advanced, metastatic cancer.

"If you are a patient with early stage breast cancer and you hear about a great new drug for metastatic breast cancer, it's probably not going to be available to you under the current model for a decade," said Dr. Tatiana Prowell, a cancer specialist at Johns Hopkins in Baltimore and a medical officer for the FDA.

Prowell and Pazdur wrote about the guidelines in the current New England Journal of Medicine.

PART OF PUSH AT FDA

The change is part of a push at the FDA to spur innovation and get more effective drugs to the patients who need them.

"We really do want to be very forward-leading," FDA Commissioner Dr. Margaret Hamburg said in a briefing with Reuters on Thursday.

"We understand the importance of taking an advance in science as quickly as we can to the patients who need them, always being mindful of our responsibility of looking at the issues of safety and efficacy."

FDA is taking a risk with the new strategy, which will expose women with much earlier-stage disease to potential safety issues of new medications.

Traditionally, by the time new drugs are tried on women with earlier-stage disease they have been tested in thousands of women with much more advanced disease, for whom the risk of trying the drug is balanced with its potential for prolonging their lives.

To help mitigate these risks, the new guidance document will apply to women with an especially deadly form of breast cancer called triple-negative breast cancers.

Most breast tumors are called estrogen-receptor positive, because they are fueled by the hormone estrogen. About 20 percent are HER2-positive, because a protein called HER2 is involved. A third type is driven by the hormone progesterone.

These types of cancer have good treatments.

Then there are basal-like or triple-negative tumors, so named because they lack estrogen, progesterone or HER2 receptors needed for most breast cancer drugs to work.

Triple-negative breast cancers tend to grow and spread more quickly than most other types of breast cancer, and they occur more often in younger women and in African-American women.

"NOT FOR EVERY PATIENT"

"This is not for every patient. It should be for patients that are most likely to benefit and also those that are at highest risk of having a recurrence of the disease," Pazdur said.

Trials would be structured so that women are treated with either chemotherapy or chemotherapy plus an experimental drug for a few months prior to surgery.

If no sign of the cancer is found during surgery, the woman has achieved a complete pathological response. Researchers would then compare response rates in the two groups, and if the drug has helped cure significantly more cancers, it would be given a provisional type of approval called accelerated approval.

Drug companies would continue to follow women for several years to see if their cancers come back, and if women continue to be disease-free, companies could get full approval for the treatment. FDA is accepting comments about its draft plan through the end of July.

Reaction so far has been positive.

"We clearly need new approaches to getting stuff to the early-disease setting," said Dr. George Sledge, co-director of breast cancer program at the Indiana University Simon Cancer Center and immediate past president of ASCO.

FASTER ACCESS TO NEW DRUGS?

He said the new approach is not a perfect solution to the problem, but it's a good first step.

"It recognizes that we want to try to move drugs up front into an early disease setting as soon as possible - not wait until the patient is beat up and out of chances and out of hope before we throw a drug at the patient and hope it does something."

Leerink Swann analyst Howard Liang said the new approach could potentially speed access to several breast cancer drugs in development, including treatments by Roche, GlaxoSmithKline , Celgene Corp, Onyx Pharmaceuticals, Bayer, Boehringer Ingelheim, Eli Lilly and Co, Eisai Co Ltd and Puma Biotechnology Inc.

FDA is basing much of its plan on a clinical trial known as I-SPY 2, a Phase II breast cancer trial being conducted by the Biomarkers Consortium, a public-private partnership run through the Foundation for the National Institutes of Health.

The trial, being run by Dr. Laura Esserman at the University of California at San Francisco and Dr. Donald Berry of MD Anderson Cancer Center in Houston, relies on specific genetic signatures or biomarkers in tumors to select those who might benefit from testing with this new approach.

The trial uses an approach called adaptive design, in which researchers continuously monitor which patients are responding to drugs and try to give drugs to those most likely to respond.

"If a therapy is doing well in a particular subset of patients, it gets a higher probability of being assigned to a patient in that subset," Berry said.

The design allows the team to test new treatments with far fewer participants and in half the normal time, cutting back on the costs of developing treatments.

Hamburg said she is meeting with the board at the ASCO cancer meeting this week to discuss this and other approaches to innovative clinical trial design.

"We feel more powerfully than ever before that how the studies are structured from the get-go really matters for our ability to do our job in an effective and efficient manner," she said.

"We really do want to be part of the scientific discussions early on to help shape how research is done."

Breast cancer is the second-leading cause of cancer death in women, exceeded only by lung cancer, according to the American Cancer Society.

Mubarak verdict adds to tension before Egypt vote

CAIRO (Reuters) - Egyptian pro-democracy campaigners called for a new uprising on Sunday, enraged that a court had spared former leader Hosni Mubarak his life over the killing of protesters during the street revolt that ended his three-decade rule.

In the first judicial reckoning of a leader toppled in last year's Arab spring uprisings, Mubarak was handed a life prison sentence. His sons were found innocent of corruption charges and senior policemen were acquitted.

Thousands took to the streets for protests that went on through the night in Cairo's Tahrir Square and in other cities, adding to political tension building since Mubarak's last prime minister made it through to a presidential election run-off.

Many took the verdicts as proof the Mubarak clan still holds sway as Egypt prepares for the vote on June 16 and 17, billed as the final stage of an army-led transition to democracy.

"This was not a fair verdict and there is mass rejection of the judge's ruling," said one protester, Amr Magdy. "Tahrir will fill up again with protesters. In Egypt the only way you can get any justice is by protesting because all the institutions are still controlled by Mubarak figures."

Many of the young liberal and left-wing revolutionaries who led last year's uprising were dismayed when their own candidates lost the first round of a presidential election last month.

The run-off will pit Mubarak's last prime minister Ahmed Shafiq, who holds Mubarak as a role model, against the candidate of the socially conservative Muslim Brotherhood, Mohamed Mursi.

Suspicion is widespread that the military, led by Mubarak's old defence minister, will still wield heavy influence after the new president takes office. The electoral success of Shafiq, a former air force commander, has deepened those fears.

Egypt has been led by army officers since the king was overthrown in 1952.

On Sunday morning, a few hundred protesters gathered in Tahrir Square -- focal point of the January 2011 revolt that brought down longtime U.S. ally Mubarak - vowing to stay until there was justice for those killed in the uprising.

"Yesterday people were united like in the early days of the revolution. I felt the revolution is returning," 46-year-old engineer Osama Awad in Tahrir said.

He said Mubarak must be tried again because key evidence had been concealed and Egyptians must unite against Shafiq.

"When Mursi wins, we can re-try Mubarak and the old regime," Awad said.

CLOSED-DOOR MEETING

Dozens of young men ransacked Shafiq's campaign office in Fayoum south of Cairo overnight, the second such attack in recent days, state news website al-Ahram reported. A Shafiq campaigner in Cairo said he was not aware of the attack.

Footage posted on Al-Ahram's website showed youths destroying and burning Shafiq's pictures and banners and others chanting: "Fayoum says Ahmed Shafiq is feloul," an Arabic word used to refer to remnants of the Mubarak era.

Shafiq has taken a tough stance on law and order, appealing to many Egyptians tired of political chaos and insecurity that have damaged the economy and worsened poverty. Critics say he also has the backing of the powerful army.

Leftist Hamdeen Sabahy, who failed to progress to the election run-off, joined thousands of protesters in Tahrir late on Saturday. The Brotherhood's Mursi also toured the square.

Mursi, who has been struggling to rally the support of candidates defeated in the first-round, met with Sabahy and another defeated candidate, Islamist Abdel Moneim Abol Fotouh, on Saturday night for a closed-door meeting, according to people who attended the gathering.

Sabahy and Abol Fotouh, who was ejected from the Brotherhood last year and has campaigned on a more moderate political platform, came third and fourth in the May 23-24 vote and have refused to throw their weight behind Mursi.

"The situation now is deadlocked but one scenario would be to stop the second round from happening," said a political activist who witnessed the meeting.

"We plan to call for marches on Monday, Wednesday and a big million-man march on Friday," the activist said.

MUBARAK "HEALTH CRISIS"

Poverty, police brutality and anger at corruption drove the 18-day revolt that unseated Mubarak. Around 850 people were killed as the security forces tried to re-assert control over the country of 82 million people.

When the fallen leader was first wheeled into a courtroom on a hospital trolley last year, it sent shockwaves through the Arab world, where autocrats have long held sway.

The defendants denied charges that ranged from the killing of the protesters to corruption and abuse of power.

State television said Mubarak suffered a "health crisis" after the verdict when he was flown to Cairo's Tora prison, where he was admitted to a hospital facility. He had been held at a luxurious military-run hospital during the 10-month trial.

Former Interior Minister Habib al-Adli was also given life in prison. Mubarak's sons Alaa and Gamal had corruption charges quashed, but stay in jail over another case.

The judge acquitted the other senior security officials for lack of evidence, a decision that worried lawyers for victims' families who said that could help Mubarak win any appeal.

"If my son had been killed and I heard this ruling, I would want to go out and avenge him," said lawyer Sayed Nosseir, who was protesting in Tahrir.

Scots guardsmen cupcake toppers, union jacks on toothpicks, ice queen scoops. Bunting: enough to string together the old empire. Royal photos on thimbles, spoons, tea cups, shot glasses, egg cups, tea towels ? the usual tat, as the Brits say. Jubilee thongs? Yes.

Then there?s the cleverness contest surrounding the royal saying, ?Keep Calm and Carry On.? Written on a women's compact: ?Keep Calm and Powder On.?
Comparatively, Google?s doodle is an understated affair. The letter O?s are diamonds, symbolic of the 60th (diamond) anniversary on the throne. The E sits as a jewel in the queen?s crown. The queen appears in two-dimensional contour; her beloved corgis, however, look ready to leap off the page.

The playful riffs on royal themes reflect the generally favorable mood toward the royals of late.

A YouGov poll last month found 67 percent agreeing the monarchy is good for Britain, and 86 percent approve of Queen Elizabeth. This is up from the days when her children's divorces dominated the headlines and her slowness to show emotion over the death of Princess Diana frustrated many.

Still, the public mood is not really one of royal reverence. Britons are in party mode at the start of a long weekend ? Monday and Tuesday being holidays as part of the four-day official jubilee celebration. Many Londoners took the chance to flee on summer vacations, as foreign and British tourists crowd into the city for events that include Sunday?s royal family flotilla on the Thames and Tuesday?s carriage procession.

Among the street art for the crowds will be the construction Sunday of a 94-sq.-ft. portrait of Queen Elizabeth made from 3,120 squares of cake ? one for each week of her reign.

German-born baker Gerhard Jenne came up with the idea. Mr. Jenne dipped each piece of lemon cake into one of 24 frosting colors, turning each into a pixel of this icing illustration.

Jenne?s a fan of the queen, but not so much so that he feels the need to become her subject even 30 years after moving to London. He doesn?t expect to meet her on Sunday, when he publicly arranges the cake squares into the portrait.

?It?s very interesting how the queen has changed. Even from 30 years ago, [the royals] are much closer to the people now,? he says. ?You almost feel like she might come see you?. There could just be a 1 in 40 million chance.?

After spending hundreds of hours on the cake, he expects it to be gobbled up within 20 minutes when it?s opened up to the crowds.

Some customers in his Konditor & Cook bakeries have asked him if it?s proper for an image of Her Highness to be eaten.

?I just said yes, it?s fine by me. At the end of the day, it?s a cake, it?s a bit of fun. And she seems like a humorous person.?

Oncothyreon Announces Presentation of PX-866 Clinical Data at ...

Oncothyreon (NASDAQ: ONTY) announced Saturday that data from two clinical trials of PX-866, a pan-isoform phosphatidylinositol-3-kinase inhibitor, were presented today at the American Society of Clinical Oncology meeting in Chicago. Oncothyreon also provided an update on the status of its ongoing Phase 2 development program for PX-866.

Phase 1 Trial of PX-866 in Combination with Docetaxel
Data from the Phase 1 portion of the ongoing Phase 1/2 of PX-866 in combination with the chemotherapeutic agent docetaxel (Taxotere?) were presented by Antonio Jimeno, M.D., Ph.D., of the University of Colorado Cancer Center, Aurora, Colorado. The trial enrolled 43 patients with advanced cancer for whom docetaxel was considered standard of care. Patients were treated at three different dose levels of PX-866 administered daily in combination with the standard dose of docetaxel (75 mg/m2) administered once every three weeks. No dose-limiting toxicities were identified, and the recommended Phase 2 daily dose of PX-866 to be given in combination with docetaxel was determined to be the same as the single agent daily maximum tolerated dose of 8 mg.

The combination of PX-866 and docetaxel was generally well-tolerated, with most adverse events being mild to moderate in severity. The safety profile for combination treatment was consistent with that for either drug administered alone. Combination treatment had no impact on the pharmacokinetic profile of either drug.

Thirty-three patients were evaluable for response, defined as having undergone at least two cycles of therapy and a follow-up tumor assessment. In these patients, the disease control rate (partial response or stable disease) was 85 percent (28/33) after two cycles of therapy and 58 percent (19/33) after four cycles. Ten patients were on study for more than 200 days, five of whom remained on therapy at the time of the report. Nine patients with stable disease or better discontinued docetaxel after four or more cycles and continued on single agent PX-866, including all five remaining on therapy. No statistically significant differences in progression-free survival were identified for patients with PIK3CA mutations or PTEN deficiency.

Phase 2 Trial of PX-866 in Patients with Glioblastoma Multiforme
Interim data from an open-label single-arm Phase 2 trial of PX-866 in patients with glioblastoma multiforme, a type of brain cancer, were presented by Marshall W. Pitz, M.D., of CancerCare Manitoba, University of Manitoba, Winnipeg, Manitoba. The Phase 2 trial is being conducted at 7 Canadian centers by the NCIC Clinical Trials Group, Queen's University in Kingston, Canada, in patients whose brain tumor is in first relapse during or following primary therapy. The trial has a two-stage design, with progression to the second stage dependent upon achieving a pre-specified endpoint of response or lack of early progression.

Seventeen patients were enrolled in the first stage of the trial and were included in this presentation. PX-866 was well-tolerated in the study, with the most common adverse events being diarrhea and reversible liver enzyme elevation. All patients were evaluable for response, with one patient having a partial response and six patients with stable disease, three of whom remained on therapy at the time of the report. These efficacy data met the pre-specified endpoint, and the trial is currently enrolling a planned additional 15 patients in the second stage.

PX-866 Phase 2 Program Update
In addition to the above mentioned trial in patients with GBM, the NCIC CTG is also conducting a Phase 2 trial in up to 40 patients with metastatic or recurrent castration resistant prostate cancer who have not received prior chemotherapy. The primary endpoint of this single-arm screening trial is the proportion of patients with lack of disease progression at 12 weeks from the initiation of therapy with PX-866. The trial has a two-stage design, with progression to the second stage dependent upon achieving a pre-specified endpoint of lack of early progression. The trial has completed enrollment in the first stage of the study and the interim analysis is currently pending.

The Phase 1/2 trial of PX-866 in combination with docetaxel mentioned above has advanced to the Phase 2 portion, which is an open-label, randomized evaluation of the antitumor activity and safety of PX-866 administered at the recommended daily dose in combination with docetaxel, versus docetaxel alone, in two groups of patients. Group 1 is enrolling patients with non-small cell lung cancer receiving second or third line treatment. Group 2 is enrolling patients with locally advanced, recurrent or metastatic squamous cell carcinoma of the head and neck after failure of prior therapy. The two groups will be randomized and evaluated independently. Enrollment of a planned 88 patients in the NSCLC group is currently expected to be completed this month. Additional centers are being added to the SCCHN arm of the trial, with a goal to complete enrollment of 82 patients in the first half of 2013.

Oncothyreon has also advanced to the Phase 2 portion of a Phase 1/2 trial of PX-866 in combination with the chimeric monoclonal antibody cetuximab (Erbitux?), which is an open-label, randomized evaluation of the antitumor activity and safety of PX-866 administered at the recommended daily dose in combination with cetuximab, versus cetuximab alone, in two groups of patients. Group 1 is enrolling patients with metastatic colorectal cancer who have a history of progression or recurrence following prior treatment with irinotecan and oxaliplatin containing regimens or who are intolerant of irinotecan. Over 50 of a planned 80 patients are currently enrolled in this arm of the trial, with enrollment currently expected to be completed in the fourth quarter of 2012. Group 2 is currently enrolling patients with incurable progressive, recurrent or metastatic SCCHN. The two groups will be randomized and evaluated independently.

Oncothyreon also recently announced the initiation of a Phase 1/2 trial of PX-866 in combination with vemurafenib (Zelboraf?). The Phase 1 portion of this trial will evaluate the safety and tolerability of PX-866 in combination with twice daily oral administration of vemurafenib in up to 36 patients with any BRAF-mutant cancer. The Phase 2 portion of the trial will compare the anti-tumor activity and safety of PX-866 and vemurafenib at the doses recommended from Phase 1 with vemurafenib alone administered at the approved dose. This randomized Phase 2 trial is expected to enroll 110 patients with advanced BRAF-mutant melanoma and has a primary endpoint of progression-free survival. This Phase 1/2 trial is being conducted in collaboration with the Melanoma Research Foundation Breakthrough Consortium.

Send money now! Candidates compete for online cash

(AP) ? Republican strategist "Karl Rove and his allies have taken the gloves off" in Ohio. Send money to stop them.

No, wait. "Hollywood-liberal-elites are trying to hijack a Senate seat in Missouri." Funds needed now to prevent it.

These aren't letters home from distraught relatives or friends. They are part of a ceaseless competition for campaign cash in the email era, from the race for the White House to Congress and local office.

The stakes are high, measured in the hundreds of millions of dollars every election cycle. Precisely how much is not known, since the Federal Election Commission does not require federal candidates to tally donations raised via email or websites separately from those made in response to traditional mail, phone banks or candidate calls.

In an age of multi-tasking, getting attention fast is critical.

President Barack Obama entered small-dollar donors into a lottery with a chance to have lunch with him last fall. In a follow-up, the prize is dinner with him and former President Bill Clinton.

The idea seems to be catching on. Mitt Romney's campaign is raising funds by giving contributors a chance to be one of four picked to "sit down for a bite to eat" with the Republican presidential contender and his wife, Ann.

Some online appeals include video, like one Sen. Debbie Stabenow, D-Mich., included of her rival, Pete Hoekstra, saying he favored drilling for oil in the Great Lakes laterally from onshore platforms.

Others seek a signature on an online petition, an act meant to create a sense of empowerment in the signer, and one that leads quickly to a request for funds.

Opponents of the recall of Wisconsin Gov. Scott Walker often send email requests for money that include a list of names of "Great Patriots" and the amounts they have donated. The hope is that others will join.

"It's like a two-minute elevator pitch," said Taryn Rosenkranz, whose company, New Blue Interactive, works for Democratic candidates and causes. "You don't have very much time before you've lost the reader."

Messages delivered digitally are "faster and more agile than TV, direct mail or phones. You can initiate a fundraising campaign the day a news story hits or something of note occurs in the political environment," said Ben Olson, director of online services for the Republican-aligned firm Arena Communications.

Even preview lines ? those short phrases that summarize items in an email inbox ? are viewed as critically important. "Nasty, vindictive and liberal to boot!" read one recently, practically begging to be opened. "Exclusive: We want you to be the first to see this," confided another mass email.

Technology lets campaigns know instantly how much money is coming in the door in response to the latest pitch. "A lot of times what you can do is put out two or three different versions and put them out to different demographics and sometimes through different websites," said Steve McMahon, a Democratic political consultant.

Increasingly, campaigns use Facebook and other social media websites to raise money. Erik Nilsson, vice president at CMDI, a Republican-aligned firm, claims credit on the company's website for showing that online fundraising yields "can be increased by 52 percent by engaging donors through social networks."

In an interview, Nilsson said, "Friends asking friends are more likely to get a donation and when those donations come in ... they come in much higher."

The next frontier may be donation by text message, which is currently banned.

Among the obstacles is a long lag between the time a donation is made and when it is transferred to the campaign by the mobile company. Also, a text donation to a charity, for example, provides a donor's cellphone number, but not name, address and occupation, information the Federal Election Commission requires to ensure a contribution is legal.

Lawyers representing a pair of consulting firms, one allied with each of the two major political parties, have recommended steps to overcome the difficulties and last month asked the FEC to permit donations by texting.

None of the technical considerations is readily apparent to the potential donor, left to sift through competing appeals.

No event or issue, it seems, is too minor to trigger an urgent and/or outraged request for contributions.

The campaign of Sen. Sherrod Brown wants money because Rove and his allies "have taken the gloves off" and are attacking the Ohio Democrat. "If we can't hit the million-dollar goal for our No Fear Fund, we'll get buried before the summer even starts," said a recent email.

Sarah Steelman, a Republican running for the Senate in Missouri, warned recently that Democratic incumbent Claire McCaskill "has a series of liberal celebrities who are funding her campaign ... defeat Hollywood's Third Senator," it says. Photos of Danny DeVito, Susan Sarandon, Kate Capshaw and Steven Spielberg are included, superimposed on the iconic Hollywood sign in the hills of Los Angeles.

In recent days, Florida Democrats claimed they had registered 10 percent more voters than Republicans and pleaded, "Contribute today and help us keep the momentum."

South Carolina Gov. Nikki Haley's political organization emailed supporters about her new book, "Can't Is Not an Option."

"If you donate $50 or more to help me continue our fight, you will receive a limited edition personalized copy," she wrote.

Tommy Thompson, a Republican contender for the Senate in Wisconsin, told his email recipients somewhat breathlessly, "We just received our first shipment of yard signs." Anyone interested in having one could stop by the office. If not, they could join the campaign as a volunteer.

Either way, a $20 donation means "we can keep buying more signs."

Make Your Next Home Improvement Job Successful

This article has been viewed 13 times.

There seems to be an air of anticipation and perhaps breath-holding when you begin an important home improvement project. Of course, you will have a wide range of projects, with some more complicated than others. Your level of experience is important but even people who have extensive experience can have problems. This is because you rarely know what will happen until you get to work. Frequently, you will find you have no choice except to roll with the punches and go with the flow. But that is all right, and here are some helpful suggestions are ideas for you.

All homeowners would like to have more space to live in and enjoy their environment. You will first need to analyze what?s in your home first. Your basement, for example, could be transformed into something else. As long as your attic isn?t brimming full of stuff, you could always work with it to create something new.

You could also consider building an addition to your property, if you have enough cash and the zoning in your area allows for it. Some home owners have made two story houses out of ranch houses. The only real limit is your imagination and the resources available to you. If you need some help to come up with ideas, then you could use some design software. Consistent or chronic power problems could be a sign that you are putting too much of a strain on your breakers. Another possibility could be with a degraded circuit breaker either individually or elsewhere in your system. You might also want to check and see if any of your household appliances are in the process of failing. For projects like this though an electrician might be your best choice. It can be simple to identify the problem if the same breaker is the one that always seems to be tripping. Safety should be your primary concern though.

Make sure you know about and are prepared to deal with any potentially hazardous materials in and around your home. Don?t even think about dealing with hazardous materials yourself; call in professionals who are trained to handle it. Dangerous materials are required, by law, to be disposed of in very specific ways. If you do it yourself you could be exposing your family to health and legal troubles. The large number of older homes in the US means that this problem isn?t going to slow anytime soon.

Home improvements involving automation attract us a lot because the results are just fun to have. Your imagination and creativity are the only obstacles to achieve what you want, especially since you can program virtually any kind of function.

Sheep free of Chernobyl controls

Restrictions covering sheep movements after the Chernobyl nuclear disaster have finally been lifted from all farms in England and Wales after 26 years.

After the 1986 disaster, the Food Standards Agency (FSA) placed controls on 9,800 UK farms, but these were gradually removed.

The final eight in Cumbria and the last 327 in Wales are now free of them.

Adam Briggs, from the NFU, said it meant an end to the "sorry situation" of the Chernobyl legacy.

As a result of the explosion, radioactive particles became locked in upland peat and accumulated in grazing sheep.

'Eat with confidence'

Under the "mark and release" restrictions, the animals had to be tested for levels of Caesium-137 prior to being moved down from the fells for sales.

Farmers were paid ?1.30 per animal, to compensate for the cost of holding them before monitoring.

The controls were lifted in Northern Ireland in 2000 and in Scotland in 2010.

Adam Briggs, policy advisor for the NFU in the North West, said: "I've spoken to a number of farmers in the area and they are all very happy with the decision.

"It gives them a bit of flexibility of when they can market the stock.

"The main thing is that research shows the meat is safe.

"People can now eat Cumbrian lamb with confidence."

New jobless claims rise for 4th straight week

By msnbc.com staff and news wires

New claims for unemployment benefits rose again in the latest week, raising worries that?labor market growth was stalling.

The Labor Department reported Thursday that seasonally-adjusted claims increased by 10,000 to 383,300. It was the fourth week in a row that claims have risen.

The four-week moving average, considered a more accurate gauge of labor market conditions,?increased by 3,750 to 374,500. The prior week's figure was revised up to 373,000 from the previously reported 370,000. Economists polled by Reuters had forecast claims unchanged last week.

Claims have now risen in seven of the last eight weeks. Most of those increases were marginal and the overall level of claims has held at levels consistent with a modest recovery in the labor market.

But the steady increase could add to the concerns raised by April's disappointing 115,000 gain in nonfarm payrolls. A Labor Department report due on Friday is expected to show employers added 150,000 jobs in May.

The four-week moving average for new claims, a measure of labor market trends, increased 3,750 to 374,500.

A Labor Department official said there was nothing unusual in the data but that results for five states, including California, had been estimated.?
